3.Product introduction
Lemairamin or Wgx50 is a natural compound derived from Sichuan pepper, which has
shown promising effects in the prevention and treatment of Alzheimer's. It has been
found to inhibit Aβ-induced neuronal apoptosis, reduce neuronal calcium toxicity,
and decrease the accumulation of Aβ oligomers in the cerebral cortex.
Additionally, wgx50 has been shown to improve the cognitive abilities of mice.
Further investigation using atomic force microscopy (AFM) revealed that wgx50 can
directly inhibit Aβ oligomers. Co-incubation of Aβ42 peptide with wgx50 prevented
Aβ fibril growth, disassembled oligomers, and only a few fibrils remained.
Molecular docking and MD simulations showed that wgx50 binds to hydrophobic grooves
on the surface of Aβ protofibril and destabilizes the structure, disrupting the
D23-K28 salt bridges, and partially opening the two tightly compacted β-sheets.
This indicates that wgx50 has the potential to prevent the intermolecular protein-
protein assembly of amyloid.
Overall, the findings suggest that wgx50 has potential as a therapeutic agent for
Alzheimer's and may provide a new avenue for the development of drugs that target
Aβ oligomers. However, further studies are needed to investigate its efficacy and
safety in humans.
